Actually not necessarily, or should i say not per say.

Most Intel chipset-based boards support asyncronous dual channel (e.g. 3x1 GB will run in that mode).
There's also flex mode:

I do know running 3 matching dimms does indeed work in asyncronous dual channel on my mobo.

Anyway, realistically though, RAM is cheap enough that'd i'd recommend buying matching pairs anyway, even if running a 32-bit OS that can't fully utilize all of it.
